**Conatumumab + mFOLFOX6** is a combination therapy of conatumumab, an investigational fully human monoclonal IgG1 antibody that specifically agonizes death receptor 5 (DR5), with the modified FOLFOX6 chemotherapy regimen. mFOLFOX6 consists of oxaliplatin, leucovorin (folinic acid), and fluorouracil (5-FU). This combination was investigated for the first-line treatment of metastatic colorectal cancer (mCRC) in clinical trials, but addition of conatumumab did not improve efficacy over mFOLFOX6 with placebo. Conatumumab induces apoptosis of tumor cells via DR5 activation. mFOLFOX6 acts through DNA crosslinking (oxaliplatin), inhibition of thymidylate synthase (fluorouracil), and enhancement of 5-FU cytotoxicity (leucovorin)[1][2][3][6].
