Overview

Concizumab is a humanized IgG4 monoclonal antibody that targets the Kunitz-2 domain of tissue factor pathway inhibitor (TFPI), a key regulator of the coagulation cascade. By inhibiting TFPI, concizumab enhances the production of factor Xa via the extrinsic clotting pathway, thereby improving thrombin generation and clot formation. This mechanism is effective even in patients with hemophilia A or B who have developed inhibitors to standard clotting factor replacement therapy, as it bypasses the intrinsic pathway where factors VIII and IX act. Concizumab is indicated for routine prophylaxis to prevent or reduce bleeding episodes in patients aged 12 years and older with hemophilia A (congenital factor VIII deficiency) with FVIII inhibitors or hemophilia B (congenital factor IX deficiency) with FIX inhibitors[1][4][5][7]. The drug is administered subcutaneously once daily[2][3].
