Conteltinib is a potent oral multi-kinase inhibitor primarily targeting anaplastic lymphoma kinase (ALK), focal adhesion kinase (FAK), and proline-rich tyrosine kinase 2 (Pyk2). It exhibits significant inhibitory activity against FAK with an IC50 of 1.6 nM. Conteltinib has demonstrated anti-tumor activity in preclinical and clinical studies, particularly in advanced ALK-positive non-small cell lung cancer (NSCLC) and pancreatic cancer. The drug also acts as an IGF type 1 receptor antagonist. Developed as a new molecular entity by Centaurus Biopharma and partners, it is currently in phase III clinical trials for NSCLC and pancreatic cancer[1][2][3][6][8].
