Contineum Therapeutics is developing a small molecule inhibitor targeting the calpain-calpastatin proteolytic system. Calpains are a family of calcium-dependent cysteine proteases that play critical roles in cellular signaling, protein degradation, and apoptosis. The activity of calpains is tightly regulated by calpastatin, their endogenous inhibitor; however, pathological overactivation of calpains has been linked to various conditions, including neurodegeneration, muscular dystrophy, and tissue injury. Contineum's program aims to modulate this system to restore proteolytic balance. The candidate is currently in the preclinical stage of development, with the primary therapeutic indication remaining undisclosed.
