Overview

COPAD is a multi-agent chemotherapy regimen used primarily in the treatment of pediatric and young adult mature B-cell malignancies, including Burkitt lymphoma/leukemia, diffuse large B-cell lymphoma (DLBCL), mediastinal large B-cell lymphoma (MLBCL), and B-cell acute lymphoblastic leukemia (B-ALL). Developed and studied by St. Jude Children's Research Hospital as part of the 'Mature B-Cell Lymphoma and Leukemia Study III' (SJBC3), the regimen is an acronym for its active components: cyclophosphamide, vincristine (Oncovin), prednisone (or prednisolone), doxorubicin (Adriamycin), and G-CSF (filgrastim or pegfilgrastim). COPAD is a combination cytotoxic chemotherapy regimen that targets rapidly dividing cancer cells through multiple mechanisms, including DNA alkylation, microtubule inhibition, and topoisomerase II inhibition. It was originally derived from the LMB-96 (Lymphome Malins de Burkitt) protocol and is specifically utilized for lower-risk (Group A) patients in risk-adapted clinical trials.
