Overview

Copanlisib is a small molecule, intravenous kinase inhibitor used primarily for the treatment of adults with relapsed follicular lymphoma who have received at least two prior systemic therapies. It acts as a selective pan-Class I phosphoinositide 3-kinase (PI3K) inhibitor, with preferential activity against the alpha (PI3K-α) and delta (PI3K-δ) isoforms that are commonly overexpressed in malignant B cells. By inhibiting PI3K signaling pathways—including those downstream of the B-cell receptor—copanlisib disrupts cell proliferation and survival signals, induces apoptosis, and inhibits chemotaxis and NFκB signaling in lymphoma cell lines. The drug was developed by Bayer and was granted accelerated FDA approval in September 2017 for follicular lymphoma. Copanlisib is administered intravenously.
