Overview

The combination of copanlisib and eribulin is an investigational therapy for advanced or metastatic triple-negative breast cancer (TNBC). Copanlisib is a pan-class I PI3K inhibitor that blocks the PI3K pathway, which is frequently activated in TNBC and associated with chemotherapy resistance. Eribulin is a non-taxane microtubule dynamics inhibitor approved for metastatic breast cancer after prior chemotherapy. Preclinical studies show that combining these drugs leads to enhanced tumor growth inhibition, increased mitotic arrest, and greater induction of apoptosis compared to either drug alone, regardless of PI3K pathway alterations or PTEN status[1][2][5]. This combination has entered clinical trials for patients with metastatic TNBC who have received prior taxane and anthracycline therapy[2][4].
