Overview

coPMLA-Me25H75 is a partially methylated copolymer derivative of poly(β,L-malic acid) (PMLA) designed as a biodegradable and biocompatible nanoconjugate platform for tumor drug delivery. Developed by researchers at Cedars-Sinai Medical Center and the Universitat Politècnica de Catalunya, the polymer platform contains pendant carboxyl groups (with 25% of them methylated) that can be conjugated with various therapeutic agents, targeting molecules, and protective groups. It exhibits membrane-disrupting and membrane-penetrating activities, allowing direct delivery of drugs to intracellular targets by bypassing the plasma membrane. In preclinical studies, coPMLA-Me25H75 demonstrated the ability to destabilize liposome membranes at both acidic and physiological pH, and was internalized into breast cancer cells with moderate in vitro cytotoxicity.
