Overview

Copper(II) diethyldithiocarbamate (Cu(DDC)2) is a small molecule metal complex and the active anticancer metabolite formed when disulfiram (Antabuse) reacts with copper ions. It is currently being investigated as a potent antineoplastic agent across various solid tumors, including glioblastoma, pancreatic ductal adenocarcinoma, and triple-negative breast cancer. Its mechanism of action is multifaceted, involving the inhibition of the p97 (VCP) segregase pathway, induction of reactive oxygen species (ROS), and inhibition of aldehyde dehydrogenase (ALDH) activity. Due to its poor aqueous solubility and short half-life, research is focused on nanotechnology-based delivery systems, such as albumin-encapsulated nanoparticles (Alb-CuDDC), to improve bioavailability and therapeutic index.
