Overview

copper + dimercaptosuccinic acid + polyethylene glycol (Cu-DMSA-PEG) is a nanocatalytic therapeutic agent developed for the treatment of non-small cell lung cancer (NSCLC). It consists of a copper-based catalytic core stabilized by dimercaptosuccinic acid (DMSA) and surface-modified with polyethylene glycol (PEG) to enhance biocompatibility and prolong systemic circulation. The nanoparticle operates through a chemodynamic therapy (CDT) mechanism; upon internalization into the tumor microenvironment, the copper ions deplete intracellular glutathione (GSH) and catalyze a Fenton-like reaction to generate reactive oxygen species (ROS). This sustained oxidative stress induces ferroptosis, downregulates the expression of glutathione peroxidase 4 (GPX4), and inhibits DNA replication, ultimately suppressing tumor progression. While related formulations may include targeting moieties like hyaluronic acid, the PEGylated version serves as a stable, long-circulating nanocatalyst for tumor-specific oxidative damage.
