Copper-64-labeled MAB 1A3 is a radiolabeled murine monoclonal antibody designed for radioimmunotherapy and imaging of human colon cancer. The parent antibody, MAb 1A3, specifically binds to a lipid antigen that is enriched in human colon carcinoma cells. When conjugated with the chelator BAT (bromoacetamidobenzyl-tetraazacyclotetradecane-N,N',N'',N'''-tetraacetic acid) and labeled with the radioactive isotope copper-64 (Cu-64), this agent delivers targeted radiation to tumor sites expressing the antigen recognized by MAb 1A3. Preclinical studies in hamsters bearing GW39 human colorectal carcinoma xenografts have demonstrated both tumor localization and therapeutic efficacy of this agent at specific dose ranges[1][3][7]. The mechanism involves selective binding to tumor-associated antigens followed by localized delivery of beta radiation from Cu-64 decay.
