The COPZ1-targeting coatomer inhibitor is a preclinical small-molecule drug being developed by Senex Biotechnology for the treatment of various cancers. The drug's mechanism of action involves the inhibition of COPZ1 (coatomer protein complex subunit zeta-1), a component of the COPI coatomer complex essential for intracellular vesicle trafficking. The therapeutic rationale is based on a synthetic lethal relationship: while normal cells express both COPZ1 and its paralog COPZ2, many tumor types lose COPZ2 expression, rendering them uniquely dependent on COPZ1 for survival. Unlike many conventional therapies that target only dividing cells, this coatomer inhibitor is designed to kill both proliferating and quiescent tumor cells, potentially leading to more complete tumor eradication.
