COR-1 is a DNA-based vaccine candidate designed for both the prophylactic and therapeutic treatment of genital herpes caused by herpes simplex virus type 2 (HSV-2). Developed by Coridon Pty Ltd (a University of Queensland spin-off later acquired by Admedus), the vaccine utilizes a proprietary codon optimization technology and a mixture of ubiquitinated and non-ubiquitinated protein-encoding DNAs. It consists of plasmids encoding the HSV-2 glycoprotein D (gD2) antigen in two forms: a full-length version and a truncated version fused to ubiquitin. The ubiquitination targets the antigen for proteasomal degradation, thereby enhancing MHC class I presentation and stimulating a robust CD8+ T-cell response, while the full-length protein is intended to induce neutralizing antibodies. The vaccine is administered via intradermal injection and has been evaluated in Phase I and Phase I/IIa clinical trials to assess safety, immunogenicity, and its potential to reduce viral shedding in HSV-2 positive individuals.
