**COR-1004** is a novel investigational subcutaneously administered small interfering RNA (siRNA) therapy developed by Corsera Health for reducing low-density lipoprotein cholesterol (LDL-C) levels. It targets **proprotein convertase subtilisin/kexin type 9 (PCSK9)** via RNA interference (RNAi), knocking down PCSK9 production to increase LDL receptor availability on hepatocytes, thereby enhancing LDL-C clearance from the blood. Currently in a Phase 1 single-ascending-dose trial (NCT07229118) in healthy volunteers with LDL-C ≥100 mg/dL, evaluating safety, tolerability, pharmacokinetics, and pharmacodynamics including PCSK9 knockdown and LDL-C reduction, with durability assessed up to 12 months.[1][2][9]
