Overview
COR788 is a novel, orally available small molecule arginine gingipain inhibitor that targets the bacterial protease produced by the periodontal pathogen *Porphyromonas gingivalis*. This protease (arginine gingipain) is implicated in bacterial survival, replication, and toxicity, and has been associated with chronic oral infections and has a proposed mechanistic link to neurodegenerative diseases such as Alzheimer's disease. COR788 was developed for its potency, selectivity, pharmacologic efficacy, and pharmacokinetic properties. Both COR788 and a related compound, COR822, are brain-penetrant, and designed to serve as next-generation inhibitors that may be used alone or in combination with lysine gingipain inhibitors. The drug was selected for IND-enabling studies, including manufacturing scale-up and dose-ranging toxicology, but its development status is currently discontinued[1][3][7].
