Overview
The CORAL vaccine is a self-amplifying mRNA (samRNA) platform developed by Gritstone bio for the prevention of COVID-19. Unlike conventional mRNA vaccines, the samRNA platform utilizes a viral replicase (derived from Venezuelan equine encephalitis virus) to enable the RNA to replicate itself once inside the host cell, which allows for significantly lower dosing levels and potentially more durable antigen expression. The CORAL program is distinguished by its inclusion of both the SARS-CoV-2 spike protein and a proprietary set of highly conserved non-spike T cell epitopes (TCE) from the nucleocapsid, membrane, and non-structural proteins. This dual-antigen approach is designed to elicit a broad immune response, combining neutralizing antibodies with robust, long-lasting CD8+ T cell activity to provide protection against emerging SARS-CoV-2 variants that may evade spike-only vaccines.
