Corallopyronin A is a natural product antibiotic isolated from Corallococcus coralloides that inhibits the bacterial DNA-dependent RNA polymerase at a distinct target site, showing activity against Gram-positive bacteria and intracellular Gram-negative pathogens including Wolbachia, Chlamydia spp., Rickettsia spp., and Orientia tsutsugamushi.[9][1] It depletes Wolbachia endosymbionts essential for filarial nematode survival, resulting in macrofilaricidal effects and positioning it as a candidate for treating onchocerciasis and lymphatic filariasis; preclinical work includes solid oral formulations to improve stability and bioavailability and robust in vivo efficacy in filarial models.[9][3][7] Development is led within the German Center for Infection Research network with contributions from Helmholtz Centre for Infection Research and academic partners; GMP production and scale-up have been established with contract manufacturers to support first-in-human trials.[3]
