Overview

Cord blood-derived expanded allogeneic natural killer (NK) cells are a cell therapy product consisting of NK cells isolated from human umbilical cord blood and subsequently expanded ex vivo. These allogeneic NK cells are not genetically modified but are grown in culture to increase their numbers and enhance their cytotoxic activity. The primary mechanism of action is direct immunomodulation and cytotoxicity against tumor or abnormal target cells through recognition of stress ligands and absence of self-MHC on malignant or infected targets. Cord blood is used as the source due to its accessibility, lower risk of graft-versus-host disease, and ability to generate large numbers of functional NK cells after expansion. This therapy is being investigated primarily for hematologic malignancies (such as multiple myeloma) and solid tumors (including osteosarcoma, Ewing sarcoma, synovial sarcoma), especially in relapsed/refractory settings[2][3][4][6]. The product may be administered following lymphodepleting chemotherapy to facilitate engraftment and persistence.
