Corin 2 is a novel small molecule dual-action inhibitor designed to target the CoREST (RE1-silencing transcription factor corepressor) epigenetic complex by simultaneously inhibiting histone deacetylase 1 (HDAC1) and lysine-specific demethylase 1 (LSD1). Developed through a collaboration between Boston University and Johns Hopkins University, Corin 2 exhibits potent anti-tumor activity, particularly in human melanoma models, where it has been shown to induce tumor suppressor genes more effectively than single-target inhibitors like entinostat. Preclinical studies indicate that Corin 2 significantly slows tumor growth in melanoma mouse xenografts while sparing primary human melanocytes, suggesting a favorable therapeutic window for treating advanced malignancies. Its efficacy appears to rely on an intact CoREST complex, as genetic knockdown of complex components abolishes its superior potency relative to parent compounds.
