Overview

CORT-113083 is an investigational, orally active, small‑molecule selective antagonist of the cortisol (glucocorticoid, GR-II) receptor being developed by Corcept Therapeutics as part of its next-generation portfolio of glucocorticoid receptor modulators. Preclinical studies demonstrated that CORT-113083, along with the related compound CORT-112716, prevented olanzapine‑induced weight gain and favorably modulated metabolic parameters in animal models, consistent with blockade of glucocorticoid signaling without appreciable activity at the progesterone receptor.[2][9][10] Corcept positioned CORT-113083 as a second selective GR-II antagonist advancing toward IND-enabling development for metabolic and psychiatric indications in which excess or dysregulated cortisol activity contributes to disease pathophysiology.[1][2][5][9][10]
