Overview

CORT108297 is a selective, high-affinity small molecule antagonist of the glucocorticoid receptor (GR), developed as a new molecular entity. It is designed to modulate neural and hormonal responses to stress by blocking the action of cortisol at the GR, without significant affinity for other steroid receptors. The drug has been investigated primarily for its potential to improve memory and cognition in people with mild cognitive impairment (MCI) due to Alzheimer's disease or those at risk for Alzheimer's, as well as for treating post-traumatic stress disorder (PTSD). Preclinical studies have shown efficacy in central nervous system models, and early clinical trials indicate it is well tolerated. Its mechanism distinguishes it from other PTSD treatments by directly targeting the body's stress response pathway[1][2][3][4][5][6][9].
