Overview

CORT125201 is the active metabolite of CORT125134, a small molecule under development as a selective antagonist of the **glucocorticoid receptor**. CORT125201 itself is considered pharmacologically active but is present at much lower plasma concentrations than its parent drug (typically 2% to 8% of CORT125134 levels). CORT125201 and its parent compound are being developed primarily for the treatment of **Cushing syndrome**, with the goal of providing clinical benefit through modulation of cortisol activity without significant antagonism of the progesterone receptor. This selectivity is considered an advantage over older therapies such as mifepristone. The mechanism of action for CORT125201, as with its parent, is antagonism of the glucocorticoid receptor[1].
