CORVax is a DNA-based immunotherapy platform developed by OncoSec Medical that utilizes the Cytokine Optimized Response (COR) technology. It involves the intratumoral delivery of plasmid DNA encoding interleukin-12 (IL-12), such as tavokinogene telseplasmid, followed by electroporation using the ImmunoPulse device. This approach is designed to stimulate a localized and systemic anti-tumor immune response by inducing the production of IL-12 directly within the tumor microenvironment. In the context of clinical research led by Chase Heaton, MD, specifically the Trifecta Study (NCT03823131), CORVax refers to the biological component of a combination regimen that includes the PD-1 inhibitor pembrolizumab and the IDO1 inhibitor epacadostat. This Phase II trial evaluated the efficacy of the combination in patients with unresectable, recurrent, or metastatic head and neck squamous cell carcinoma (HNSCC). While the CORVax name is also associated with OncoSec's COVID-19 vaccine candidate (CORVax-001), in oncology it identifies the DNA plasmid-based gene therapy platform.
