Overview

Corylin is a natural flavonoid compound isolated from the seeds of *Psoralea corylifolia* L. (Fabaceae). It exhibits a variety of pharmacological activities, including anti-inflammatory, antioxidant, and anti-proliferative properties. In preclinical studies, corylin has demonstrated protective effects against LPS-induced sepsis and systemic inflammation by inhibiting the production of pro-inflammatory cytokines such as TNF-α and IL-6, as well as inflammatory mediators like nitric oxide (NO) and prostaglandin E2 (PGE2). Its mechanism of action involves the suppression of NF-κB activity and the reduction of MAPK phosphorylation, which in turn downregulates the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). Additionally, corylin blocks the translocation of high mobility group box 1 (HMGB1) from the nucleus to the cytosol, a critical step in the progression of severe inflammation. It is currently being explored as a potential therapeutic candidate for the treatment of sepsis and septic shock.
