Overview

COT-143 (also known as INFEX702) is a humanized monoclonal antibody that targets the PcrV protein, a crucial component of the Type III Secretion System (T3SS) in *Pseudomonas aeruginosa*. Originally developed by Shionogi and licensed to Infex Therapeutics, COT-143 is designed to prevent the translocation of bacterial toxins into host immune cells, specifically protecting neutrophils and enhancing bacterial clearance. It is primarily being developed to reduce the frequency and severity of pulmonary exacerbations in patients with non-cystic fibrosis bronchiectasis (NCFB) who are chronically colonized with *P. aeruginosa*. The drug is currently in early-stage clinical development, following preclinical studies that demonstrated potent inhibition of *P. aeruginosa* cytotoxicity.
