Overview

COTI-2 is a clinical-stage, small molecule anti-cancer agent developed by Cotinga Pharmaceuticals (formerly Critical Outcome Technologies Inc.). It is a third-generation thiosemicarbazone that acts as an activator of misfolded mutant p53 proteins, restoring their wild-type tumor-suppressive function. Additionally, COTI-2 has been shown to inhibit the AKT signaling pathway. Identified via the proprietary CHEMSAS computational platform, the drug is being investigated in Phase 1 trials for various solid tumors, including gynecologic malignancies (ovarian, fallopian tube, primary peritoneal, endometrial, and cervical cancers) and head and neck squamous cell carcinoma, both as a monotherapy and in combination with cisplatin. It received FDA Orphan Drug Designation for ovarian cancer in 2014.
