COTI-219 is an orally bioavailable small molecule inhibitor designed to selectively target mutant forms of the KRAS protein while sparing the function of wild-type KRAS. Discovered by Cotinga Pharmaceuticals (formerly Critical Outcome Technologies Inc.) using their proprietary artificial intelligence platform, CHEMSAS®, COTI-219 aims to address the significant therapeutic gap in KRAS-mutated cancers, which account for approximately 30% of all human malignancies. Preclinical studies in rodent models have demonstrated that COTI-219 can significantly reduce tumor volume, outperforming certain standard-of-care therapies in lung cancer models. The compound is being developed for the treatment of various KRAS-driven solid tumors, including lung, colorectal, pancreatic, and thyroid cancers.
