Courage Therapeutics-MC4R-General Obesity is a selective melanocortin-4 receptor (MC4R) peptide agonist being developed for the treatment of general dietary obesity. This candidate is specifically designed to act as a GLP-1 potentiator, enhancing the weight-loss signaling of GLP-1 receptor agonists (such as semaglutide or tirzepatide) through synergistic effects in the hypothalamus. By targeting MC4R with high selectivity and potency, the drug aims to regulate energy homeostasis and body weight while avoiding MC1R-mediated pigmentation side effects. As of late 2025, the program is in the IND-enabling stage, following a four-year drug discovery campaign.
