Overview

COV155 is an investigational extended-release (ER) oral formulation of methylphenidate, a central nervous system (CNS) stimulant, developed by Mallinckrodt (specifically through its specialty generics and brands segment, SpecGx). It was designed for the treatment of attention deficit hyperactivity disorder (ADHD). The drug functions by inhibiting the reuptake of dopamine and norepinephrine through blockade of the dopamine transporter (DAT) and norepinephrine transporter (NET), thereby increasing synaptic levels of these neurotransmitters in the prefrontal cortex and basal ganglia. Mallinckrodt conducted Phase 1 clinical studies (e.g., NCT03103399) to evaluate the pharmacokinetics and the effect of food on the absorption of this specific ER formulation, likely as part of an effort to develop a product with a specific release profile or to meet updated bioequivalence standards for methylphenidate ER products.
