Overview

CoV2-06 is a potent human monoclonal antibody (mAb) that targets the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein. Discovered by researchers at the Vanderbilt University Medical Center (VUMC), it was identified from the B cells of a convalescent patient. CoV2-06 belongs to a "public" antibody lineage, specifically utilizing the VH3-53 or VH3-66 gene segments, which are commonly recruited in the human immune response to SARS-CoV-2. The antibody functions by binding to a highly conserved epitope on the RBD, thereby sterically blocking the interaction between the viral spike protein and the human angiotensin-converting enzyme 2 (ACE2) receptor. This blockade prevents the virus from attaching to and entering host cells, effectively neutralizing the pathogen. Licensed to IDBiologics (where it is designated as IDB006), CoV2-06 has been investigated as a potential therapeutic and prophylactic agent for COVID-19. While it demonstrated exceptional potency against early viral strains, its development has been challenged by the emergence of variants with mutations in the RBD that can reduce antibody binding affinity.
