Overview

CoV2-14 is a human monoclonal antibody (mAb) that targets the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein. Discovered by researchers at the Duke Human Vaccine Institute (DHVI), CoV2-14 was isolated from the B cells of a convalescent COVID-19 patient. It functions as a potent neutralizing antibody by binding to a specific epitope on the RBD that overlaps with the binding site for the human angiotensin-converting enzyme 2 (ACE2) receptor. By sterically hindering the interaction between the viral spike protein and ACE2, CoV2-14 prevents the virus from attaching to and entering host cells. Structural studies using cryo-electron microscopy (cryo-EM) have classified CoV2-14 as a Class 1 RBD-targeting antibody, which typically recognizes the RBD in its up conformation. While primarily utilized as a research tool to characterize the structural basis of viral neutralization and the impact of viral mutations, CoV2-14 has been evaluated in preclinical models for its potential as a therapeutic or prophylactic agent against COVID-19.
