CoV2 preS dTM-AF03 is a recombinant protein subunit vaccine candidate developed by Sanofi Pasteur for the prevention of COVID-19. The vaccine consists of a stabilized pre-fusion spike (S) protein antigen, CoV2 preS dTM, which is produced using a baculovirus expression system in insect cells. It is formulated with Sanofi's proprietary AF03 adjuvant, an oil-in-water emulsion containing squalene, sorbitan monooleate, macrogol cetostearyl ether, and mannitol. During Phase 1/2 clinical trials, the vaccine was evaluated as a two-dose intramuscular regimen. Although the AF03-adjuvanted formulation was tested for safety and immunogenicity, Sanofi ultimately prioritized the development of an AS03-adjuvanted version due to its superior immunogenicity profile, particularly in older adults.
