Overview

COVA208 is a bispecific HER2-targeting FynomAb, an engineered antibody–Fynomer fusion protein that simultaneously engages two distinct epitopes on the human epidermal growth factor receptor 2 (HER2) to induce potent receptor cross-linking, clustering, internalization, and degradation, leading to inhibition of downstream signaling and apoptosis in HER2-overexpressing tumor cells.[1][2][4] It is built on a pertuzumab IgG scaffold fused to a HER2-specific Fynomer (C12) recognizing a domain I epitope distinct from trastuzumab (domain IV) and pertuzumab (domain II), and demonstrated superior antitumor activity to trastuzumab and pertuzumab in multiple HER2-positive xenograft models.[2][4] Initially developed by Covagen, the molecule was advanced as a research-stage HER2 antagonist for HER2-positive cancers such as breast, gastric, and colorectal cancer, but development has been discontinued.[2][5][7][9]
