Overview

COVA322 is a first-in-class bispecific antibody fusion protein designed to simultaneously inhibit tumor necrosis factor (TNF) and interleukin-17A (IL-17A). It was developed for the treatment of inflammatory diseases such as psoriasis, rheumatoid arthritis, and psoriatic arthritis. The molecule is a FynomAb—a fusion of an anti-TNF antibody (based on adalimumab) with a Fyn SH3-derived binding protein (Fynomer) that targets IL-17A. This dual inhibition approach aimed to address limitations of current biologics by targeting two key pro-inflammatory pathways implicated in these diseases. Despite promising preclinical and early clinical data showing potent inhibition of both TNF and IL-17A in vitro and in vivo, clinical development was discontinued due to safety concerns identified during Phase 1 trials[1][3][4][5].
