Overview

**CoVac-1** is an investigational SARS-CoV-2 multi-peptide vaccine and T-cell activator developed at University Hospital Tübingen for prevention of severe COVID-19, particularly in people with congenital or acquired B-cell or antibody deficiency who may not generate adequate protective antibody responses. The product contains eight SARS-CoV-2-derived T-cell epitope peptides from multiple viral proteins, formulated with the Toll-like receptor 1/2 agonist XS15 in Montanide ISA 51 VG. Subcutaneous administration is intended to induce broad, durable, multifunctional SARS-CoV-2-specific CD4-positive T-helper 1 and CD8-positive T-cell responses that are less dependent on spike antigen recognition and reported to remain active against evaluated viral variants. Phase I evaluation in healthy adults and Phase I/II evaluation in patients with B-cell deficiency reported favorable tolerability and substantial vaccine-specific T-cell responses.
