CoVaccine HT is an oil-in-water emulsion adjuvant composed of hydrophobic, negatively-charged sucrose fatty acid sulphate esters and squalane. It is designed to enhance the immune response to vaccines by promoting both humoral and cellular immunity. The adjuvant has been shown to induce rapid, robust, and balanced immune responses—including increased antibody titers, immunoglobulin subclass diversity, interferon gamma production, and higher neutralizing antibody levels—when used with recombinant protein subunit vaccines such as those targeting SARS-CoV-2. Its mechanism of action involves immunostimulation that may be partially mediated through TLR4 signaling pathways. Originally developed by Pepscan Holding NV (and later Protherics), it has been evaluated in clinical trials for use in therapeutic cancer vaccines (e.g., prostate cancer) as well as infectious disease vaccines including COVID-19[1][3][5][7][8].
