The covalent heterobivalent inhibitor (cHBI) is a first-in-class small molecule inhibitor being developed by Artin Immunology for the prevention of peanut allergic reactions. The drug functions by covalently and irreversibly binding to peanut allergen-specific IgE antibodies, specifically targeting those that recognize immunodominant epitopes on the Ara h 2 and Ara h 6 allergens. By permanently blocking these IgE antibodies, cHBI prevents them from recognizing peanut proteins and triggering the mast cell degranulation that leads to anaphylaxis. Preclinical studies in humanized mouse models have demonstrated that a single dose can provide protection for over two weeks and can even prevent fatal anaphylaxis when administered shortly after symptom onset. The technology was developed through a partnership between researchers at the University of Notre Dame and the Indiana University School of Medicine.
