COVI-VAC is a live attenuated, intranasal COVID-19 vaccine developed using codon-pair deoptimization technology and deletion of the furin cleavage site in the SARS-CoV-2 spike protein. This design introduces 283 silent mutations that slow viral replication without altering viral proteins, resulting in significant attenuation and enhanced safety. Unlike spike-only vaccines, COVI-VAC presents all SARS-CoV-2 antigens in their native conformation, potentially providing broader immunity and reducing susceptibility to immune escape from emerging variants. The vaccine is administered as a single intranasal dose and has demonstrated immunogenicity, safety, minimal viral shedding, and protection against both homologous and heterologous SARS-CoV-2 challenge in preclinical models. It is currently undergoing clinical development for the prevention of COVID-19[1][2][3][5][6][7].
