COVID-eVax is a synthetic DNA vaccine candidate developed for the prevention of COVID-19. It consists of an engineered DNA plasmid encoding a secreted monomeric form of the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein. The vaccine is administered intramuscularly and delivered via electroporation, which enhances cellular uptake of the DNA. Preclinical studies demonstrated that COVID-eVax induces strong neutralizing antibody responses against SARS-CoV-2, including variants of concern, as well as robust T cell responses. In animal models such as hACE2 transgenic mice and ferrets, it conferred significant protection against viral challenge by reducing weight loss, improving pulmonary function, and lowering viral replication in target organs. The vaccine has shown a favorable safety profile in preclinical toxicity studies and entered phase I/II clinical trials in Italy[1][3][4][5][7].
