Overview

The COX1 peptide vaccine is an experimental cancer immunotherapy designed to target tumor-associated mitochondrial antigens (TAMAs). It is derived from mutated mitochondrial DNA (mtDNA) sequences, specifically targeting somatic mutations in the mitochondrial-encoded Cytochrome c oxidase subunit 1 (COX1) gene. These mutations are prevalent across various solid tumors, including renal cell, colorectal, ovarian, and breast cancers. The vaccine works by eliciting a cytotoxic T cell response against the altered mitochondrial proteins. In preclinical studies using renal cell carcinoma (RENCA) models, the COX1 peptide vaccine demonstrated therapeutic and prophylactic efficacy, suggesting that targeting mitochondrial neoantigens is a viable strategy for cancer vaccination.
