CP‑122721 is a potent and selective small molecule antagonist of the neurokinin 1 receptor (NK1R), developed by Pfizer. It was investigated for the treatment of depression, emesis (nausea and vomiting), and inflammatory diseases such as asthma and irritable bowel syndrome. The drug acts by binding with high affinity to the human NK1 receptor, thereby blocking substance P-mediated signaling pathways involved in mood regulation, emesis reflexes, pain perception, and inflammation. Preclinical studies demonstrated anxiolytic and antidepressant-like effects as well as suppression of immunocyte recruitment in models of respiratory infection[1][2][4][5].
