Overview

CP-195543 + celecoxib is a discontinued investigational combination drug that was developed for the treatment of rheumatoid arthritis. The combination consists of two small molecule agents: - **CP-195543** is a selective and potent leukotriene B4 (LTB4) receptor antagonist developed by Pfizer. It inhibits LTB4-mediated neutrophil chemotaxis and infiltration, thereby reducing inflammation. - **Celecoxib** is a selective cyclooxygenase-2 (COX-2) inhibitor, classified as a nonsteroidal anti-inflammatory drug (NSAID). It reduces pain and inflammation by inhibiting COX-2-mediated prostaglandin synthesis. The rationale for combining these agents was to target both leukotriene-driven and prostaglandin-driven inflammatory pathways in rheumatoid arthritis. However, in phase 2 clinical trials, this combination failed to demonstrate superiority over placebo, leading to discontinuation of its development[1].
