Overview

CP-BMP2 (Cell Permeable Bone Morphogenetic Protein 2) is a modified recombinant human BMP-2 engineered to enhance cell permeability, solubility, and bioavailability for improved bone regeneration. It incorporates an advanced macromolecular transduction domain (aMTD) to facilitate transfer across the plasma membrane and a solubilization domain. Mechanistically, CP-BMP2 binds to bone morphogenetic protein receptor type-1 and type-2 on target cells, activating Smad phosphorylation pathways that drive osteoblast differentiation and bone formation. Compared to traditional rhBMP-2, CP-BMP2 demonstrates superior efficacy in preclinical models of critical-size calvarial defects in mice, rabbits, and horses—showing significantly increased rates of bone formation without notable adverse effects such as ectopic ossification. The drug is being developed primarily for orthopedic indications involving accelerated or enhanced bone healing[1][6][7].
