Overview

CP-RhA01 is an investigational oral formulation of niclosamide, a small molecule mitochondrial uncoupler, being developed by Penetrium Bioscience and Hyundai Bioscience. It is designed to disrupt mitochondrial membrane potential (ΔΨm) and induce AMP-activated protein kinase (AMPK)-linked bioenergetic stress. This metabolic decoupling is intended to dismantle the pathological stromal-immune feed-forward loop (comprising activated fibroblast-like synoviocytes, remodeled extracellular matrix, and endothelial adhesion molecules) that drives tissue destruction and treatment resistance in autoimmune diseases. CP-RhA01 is under preclinical investigation for the treatment of rheumatoid arthritis (specifically persistent inflammatory refractory rheumatoid arthritis, or PIRRA), multiple sclerosis, psoriasis, and inflammatory bowel disease.
