CP101.2C8 is a fully human monoclonal IgG1 antibody targeting nucleolin, a multi-functional protein that is aberrantly expressed on the plasma membrane and in the cytoplasm of various cancer cells. Developed by CharlestonPharma in collaboration with the Medical University of South Carolina, CP101.2C8 exploits the temperature-dependent shuttling function of nucleolin to gain access to the cytoplasm of tumor cells. Once internalized, the antibody induces the destabilization of oncogene and cytokine mRNAs that contain AU-rich nucleolin binding elements in their 3'-untranslated regions (UTRs). Preclinical studies have demonstrated broad-spectrum anticancer activity against acute myeloid leukemia (AML), colon, prostate, lung, and breast cancer cells, while exhibiting negligible toxicity to normal cells that lack detectable surface nucleolin expression.
