Overview

CPA-7 is a platinum(IV)-based small molecule inhibitor of Signal Transducer and Activator of Transcription 3 (STAT3), derived from cisplatin. It functions by preventing dimerized STAT3 proteins from binding to their target DNA sequences and reducing the overall levels of phosphorylated STAT3 (pSTAT3). In the context of human papillomavirus (HPV)-associated cancers, STAT3 is a key driver of malignancy and immune evasion, often activated downstream of interleukin-23 (IL-23) signaling. CPA-7 has demonstrated the ability to induce direct tumor cell death and reverse immune suppression within the tumor microenvironment by restoring the cytotoxic and proliferative capacity of HPV-specific CD8+ and CD4+ T cells. Preclinical studies in murine models have shown that CPA-7 can lead to the complete eradication of established HPV+ tumors and the induction of durable immune memory.
