Overview

Cpd1 is a novel small molecule inhibitor of the deubiquitinase USP14 (Ubiquitin-specific protease 14), currently in preclinical development for the treatment of cisplatin-resistant ovarian carcinoma. Identified through a screening of a diversity library of approximately 11,000 compounds, Cpd1 represents a new chemical class of USP14 inhibitors with an IC50 of 18.4 μM. USP14 is a proteasome-associated enzyme that is often deregulated in ovarian cancer and contributes to the survival of tumor cells under chemotherapy stress. Cpd1 has demonstrated the ability to inhibit proteasome activity in intact cells and serves as a lead compound for further chemical optimization. The research was conducted by a collaborative team from the Fondazione IRCCS Istituto Nazionale dei Tumori and the Istituto Italiano di Tecnologia.
