CPD100PEGLi is a research-stage PEGylated liposomal formulation of CPD100, a hypoxia-activated prodrug (HAP) of the microtubule inhibitor vinblastine. Developed by Cascade Prodrug, the formulation utilizes PEGylated liposomes to encapsulate the vinblastine N-oxide prodrug, aiming to enhance its pharmacokinetic profile and tumor accumulation through the enhanced permeability and retention (EPR) effect. CPD100 is designed to remain inactive in normal, oxygenated tissues but undergoes bioreductive activation to release potent vinblastine specifically within the hypoxic microenvironment of solid tumors. This dual-targeting approach, combining liposomal delivery with hypoxia-specific activation, is intended to maximize anti-tumor efficacy while minimizing the systemic toxicities typically associated with vinca alkaloids.
