Overview
CpG 1018 is a synthetic immunostimulatory oligodeoxynucleotide (ODN) containing unmethylated cytosine-phosphate-guanosine (CpG) motifs. It acts as a toll-like receptor 9 (TLR9) agonist and is used as an adjuvant in vaccines to enhance immune responses. By activating TLR9, which is expressed on plasmacytoid dendritic cells and B cells, CpG 1018 stimulates the production of cytokines such as interferon-alpha, interferon-beta, interleukin-12, and tumor necrosis factor-alpha. This leads to increased antibody concentrations, robust T and B cell memory responses, polarization toward Th1-type immunity, and enhanced antigen presentation. The adjuvant has been clinically validated in HEPLISAV-B®, an FDA-approved hepatitis B vaccine developed by Dynavax Technologies. It is also being investigated for use in other vaccines against infectious diseases (including COVID-19) and for potential applications in cancer immunotherapy and neurodegenerative diseases like Alzheimer's disease[1][2][3][6][9].
