CpG 2216 is a synthetic Class A CpG oligodeoxynucleotide (ODN) that acts as a potent agonist of Toll-like receptor 9 (TLR9). It is structurally characterized by a chimeric backbone containing a central phosphodiester-linked CpG-containing palindromic sequence and phosphorothioate-modified poly-G ends. This unique structure promotes the formation of multimeric aggregates, which allow the molecule to be retained in the early endosomes of plasmacytoid dendritic cells (pDCs). This localization triggers robust signaling that leads to the massive production of type I interferons, particularly interferon-alpha (IFN-α), and the maturation of pDCs. CpG 2216 has been extensively studied as an immunotherapeutic agent and vaccine adjuvant, particularly for its ability to induce Tc1-biased immune responses and enhance anti-tumor immunity in contexts such as lung cancer.
